چکیده

Three major regulatory steps can be recognized in the control of ketogenesis (Fig. 1): (1) regulation of the supply of the major ketogenic substrate, plasma non-esterified fatty acids, by control of their release from adipose tissue; (2) regulation in the liver of the relative fluxes of non-esterified fatty acids through the pathways of /3-oxidation and esterification; (3) regulation in the liver of the relative fluxes of acetyl-CoA through the pathways of oxidation to CO, (via the citric acid cycle), and of ketogenesis.

CREB3L3 controls fatty acid oxidation and ketogenesis in synergy with PPARα

CREB3L3 is involved in fatty acid oxidation and ketogenesis in a mutual manner with PPARα. To evaluate relative contribution, a combination of knockout and transgenic mice was investigated. On a ketogenic-diet (KD) that highlights capability of hepatic ketogenesis, Creb3l3-/- mice exhibited reduction of expression of genes for fatty oxidation and ketogenesis comparable to Ppara-/- mice. Most of...

Ketogenesis prevents diet-induced fatty liver injury and hyperglycemia.

Nonalcoholic fatty liver disease (NAFLD) spectrum disorders affect approximately 1 billion individuals worldwide. However, the drivers of progressive steatohepatitis remain incompletely defined. Ketogenesis can dispose of much of the fat that enters the liver, and dysfunction in this pathway could promote the development of NAFLD. Here, we evaluated mice lacking mitochondrial 3-hydroxymethylglu...

The Regulation of Ketogenesis from Octanoic Acid

The regulation of ketogenesis in the rat has been studied both in uiuo and in the isolated perfused liver with octanoic acid as substrate. This fatty acid, which is not utilized directly for triglyceride synthesis, was chosen in order to investigate the events that govern the metabolism of fatty acids subsequent to the (?-oxidation process. Experiments in viuo established that for any given loa...

Mitochondrial 3-hydroxy-3-methylglutaryl-CoA synthase: a control enzyme in ketogenesis.

Cytosolic and mitochondrial 3-hydroxy-3-methylglutaryl-CoA (HMG-CoA) synthases were first recognized as different chemical entities in 1975, when they were purified and characterized by Lane's group. Since then, the two enzymes have been studied extensively, one as a control site of the cholesterol biosynthetic pathway and the other as an important control site of ketogenesis. This review descr...

The Metabolism of (- ) -0ctanoylcarnitine in Perfused Livers from Fed and Fasted Rats EVIDENCE FOR A POSSIBLE REGULATORY ROLE OF CARNITINE ACYLTRANSFERASE IN THE CONTROL OF KETOGENESIS*

In confirmation of previous findings it was shown that perfused livers from fasted rats converted oleic acid into ketone bodies far more efficiently than did livers from fed animals, whereas differences in rates of ketogenesis from octanoate were much less pronounced. However, relative rates of ketone body production from (-)-octanoylcamitine resembled those seen with oleic acid rather than tho...
